Peripheral & Central 5-HT3 Receptor Blockade: Cytotoxic chemotherapy and abdominal radiation damage intestinal enterochromaffin cells, releasing massive amounts of serotonin (5-HT). Granisetron selectively blocks 5-HT3 receptors on peripheral vagal afferent terminals in the gut and centrally in the chemoreceptor trigger zone (CTZ) of the area postrema, interrupting the vomiting reflex arc.
CYP3A4-Predominant Hepatic Metabolism (Non-CYP2D6 Dependent): Unlike ondansetron and tropisetron, Granisetron is metabolized primarily via hepatic CYP3A4 and CYP1A1 without reliance on polymorphic CYP2D6, preserving efficacy in CYP2D6 ultra-rapid metabolizers.

Directions of Use & Safety Information

Dosage & Timing: Take strictly as prescribed by your oncologist or physician. For chemotherapy or radiotherapy prophylaxis, the first dose (typically 2 mg once daily or 1 mg twice daily) is administered up to 1 hour before the start of chemotherapy or radiation (and the second 1 mg dose, if prescribed, 12 hours later).
